Wheat Genotype Database development Genotypes A collection of 222 wheat cultivars of known pedigree was assembled. Varieties were chosen to cover both CIMMYT and UK public breeding programmes, together with relevant external source varieties. The collection also included some key varieties from Australia and the USA in order that the study could be linked to similar international initiatives. Breeding data was assimilated from several sources including the JIC Germplasm database, CIMMYT's IWIS database, the National Plant Germplasm System GRIN database (http://www.ars-grin.gov/npgs/) and information supplied by individual plant breeders and a comprehensive pedigree has been prepared.SSRs Informative microsatellite loci were selected. Criteria to be satisfied included genome coverage, polymorphism levels and confidence of data scoring. SSR markers that produce a complex amplification pattern or which amplify from more than one locus were discarded where they were difficult to score with confidence. Markers were analysed on both ABI373 and ABI377 automatic sequencing instruments. Allele calling was carried out manually.The database and informatics The database is, for the time being, maintained in Excel format. A relational database is being prepared to manage this data and when ready this will be made available via the web. The new database will contain both the genotyping data and variety pedigree informationPublication and analysis.
